Mechanism of Sildenafil in the body

• The Sildenafil is the potent and active ingredient in Viagra formulations. The dose to be administered by the impotent men shall be prescribed by the physician only; it is done based on the severity of the patient. Per day the dose to be taken is either 50 mg or as prescribed by the doctor.
• The drug is taken through the oral route; it is absorbed in the gut through active diffusion, distributed in the systemic circulation through carrier mechanism, metabolized in the hepatic tissues, and excreted by the excretory organs. The peak concentration of this drug is about 4-5 hours. Therefore the erections are also seen for about 4-5 hours till the drug retains in the body. The systemic circulation enters into the smooth muscles by triggering to inhibit the phosphodiesterase enzymes.
• In men after foreplay; the nitric oxide levels increase at the penile region, which leads in the production of cGMP. The actual mechanism of cGMP is it can regulate relaxation of smooth muscles. In this context, the relaxation of corpus cavernosum is seen; thereby enhancing erections in men. Naturally the excessive cGMP levels are destroyed by the PDE enzymes. In impotent males, the PDE shall destroy the normal levels of cGMP and the erections are completely suppressed or not seen.
• Sildenafil citrate is PDE-5 enzyme inhibitor; shall allow the cGMP levels to produce indirectly in the smooth muscle region. Do not use this drug without prescription, or on friend’s advice. This drug is not suitable for the patients who are suffering with cardiac disorders.
